THIOGLYCOLLATE MEDIUM, BREWER

The thioglycollate medium, Brewer formulation, is recommended for the cultivation of anaerobes and determination of the sterility of solutions containing mercurial preservatives. Sodium thioglycollate acts as a reducing agent and also neutralises the bacteriostatic properties of mercurial compounds. Methylene blue is a redox indicator which is colourless at low Eh but turns blue on exposure to oxygen. The agar makes the medium viscous slowing down the permeation of oxygen.

PRECAUTIONS
This medium is for laboratory use only.


STORAGE
Store prepared media at 2-25C protected from direct light. Medium must not be used if there are signs of contamination or if the expiry date has passed. Store dehydrated powder in a dry place in tightly-sealed containers at 2-25C. Do not use dehydrated media if it is caked and if expiry date has passed.


DIRECTIONS
Suspend 20 grams of powder in 1 liter of purified filtered water. Mix thoroughly. Heat gently with frequent agitation and boil for one minute. Mix carefully and distribute into screw cap culture tubes or flasks. Autoclave for 15 minutes at 121C (15 psi). Tighten caps immediately after autoclaving.


PROCEDURE
This medium is straw colored, translucent, viscous which may have a blue surface due to contact with oxygen. If the medium has a diffuse blue tinge it should not be used until the oxygen has been driven off by holding in a boiling water bath for 5-10 minutes.

  1. Bring to room temperature before use.
  2. Inoculate specimens ensuring adequate dispersal of the inoculum in the broth and tighten the cap.
  3. Incubate at 35C for 24-72 hours.
  4. Examine tube daily for visual signs of growth (diffuse turbidity or individual colonies).

LIMITATIONS OF METHOD
Do not reheat tubes more than once to drive off absorbed oxygen.This medium is only a part of the identification. Other tests may be required.
