RAPPAPORT VASSILIADIS BROTH

Rappaport Vassiliadis Broth is a selective enrichment medium used for cultivation of Salmonella from feces, foods and sewage polluted water. The high concentration of magnesium chloride, malachite dye and low pH are the inhibitors.

PRECAUTIONS
This medium is for laboratory use only.


STORAGE
Store prepared media at 2-8º C protected from direct light and dehydrated powder, in a dry place, in tightly-sealed containers at 2-25º C.


SIGN OF DETERIORATION
Media should not be used if the expiry date has passed. Prepared media should not be used if there are signs of contamination or deterioration (evaporation or discoloration). Do not use dehydrated media, if it is caked.


DIRECTIONS
Suspend 26,6 g of dehydrated media in 1000 ml of purified filtered water. Heat gently with frequent agitation to dissolve medium completely. Dispense 10 ml into culture tubes and autoclave at 116º C for 15 minutes.


PROCEDURE
Prior to inoculate, the prepared media should be brought to room temperature.

  1. Inoculate tubes with specimens.
  2. Incubate at 41,5º C for 18-24 hours with loosened caps.
  3. Subculture a few drops onto selective media such a Briliant Green Agar (#1871) or XLD Agar (#1742) for Salmonella.
  4. Incubate at 35º C for 18-24 hours.

LIMITATIONS OF METHOD
Rappaport Vassiliadis Broth cannot be used for the isolation of some strains of Salmonella typhi and Salmonella choleraesuis inhibited by malachite green. This medium is only a part of the identification. Other tests may be required.
