The Stock Culture medium is used to store working control cultures at room temperature or at 2-8ºC for up to 4 weeks.

This medium is for IN VITRO diagnostic use only.


STORAGE
Store prepared media at 2-8º C protected from direct light.


SIGN OF DETERIORATION
Media should not be used if the expiry date has passed. Prepared media should not be used if there are signs of contamination or deterioration (shrinking, cracking, evaporation or discoloration).


PROCEDURE
Prior to inoculate, the prepared media should be brought to room temperature.

  1. Inoculate 0,5 ml of TSB with 1-4 colonies of a 24 hours growth of each organism to be stocked.
  2. Incubate at 35º C for 2-4 hours; than dilute a 0.5 Mc Farland Standard suspension 1:100.
  3. Dip a sterile swab into the dilution. Rotate the swab several time, pressing firmly on the inside wall of the tube above the fluid level.
  4. Transfer and dip swab into Stock Culture Medium.
  5. Store at room temperature or at 2-8oC for up to 4 weeks.

    Working strain may be restarted, as following:

  6. Incubate at 35º C for 18-24 hours.
  7. Inoculate the surface a general purpose plate and incubate at 35º C for another 24 hours.
  8. Examine for any contamination.
