Stuart's Urea Broth is a liquid differential medium by their ability to split urea, forming two molecules of ammonia by the action of the enzyme urease with a medium alkalinity. Proteus sp. give faster positive results than other microorganisms.

PRECAUTIONS
This medium is for IN VITRO diagnostic use only.


STORAGE
Store prepared media at 2-8º C protected from direct light and dehydrated powder, in a dry place, in tightly-sealed containers at 2-8º C.

 

SIGN OF DETERIORATION
Media should not be used if the expiry date has passed. Prepared media should not be used if there are signs of contamination or deterioration (evaporation or discoloration). Do not use dehydrated media if it is caked.


DIRECTIONS
Suspend 38,7 g of in 1000 ml of purified filtered water. Mix thouroughly to dissolve and sterilize by filtration through a 0,22 micron filter. DO NOT AUTOCLAVE. Dispense into sterile culture tubes.


PROCEDURE
Prior to inoculate, the prepared media should be brought to room temperature.

  1. Inoculate tubes with an overnight growth.
  2. Incubate at 35º C for 24-48 hours with loosened caps.
  3. Examine for urease activity.

LIMITATIONS OF METHOD
The high buffering system masks urease activity for organisms which exhibit a weak or delayed reaction. This medium is only a part of the identification. Other tests may be required.
