Palcam Listeria Broth is a selective enrichment medium used for isolation of Listeria sp from biological specimens, from foods and environment. Esculin hydrolysed by all Listeria sp gives blackening of the medium. Lithium chloride, polymyxin B, acriflavin HCl and ceftazidime inhibit bacterial growth.

PRECAUTIONS
This medium is for laboratory use only.


STORAGE
Store prepared media at 2-8º C protected from direct light.


SIGN OF DETERIORATION
Media should not be used if the expiry date has passed. Prepared media should not be used if there are signs of contamination or deterioration.


DIRECTIONS
Suspend 10 g in 1000 ml of purified filtered water.Heat gently to dissolve. Dispense into culture tubes. Sterilize at 121º C for 15 minutes. Cool to room temperature.


PROCEDURE
Prior to inoculate, the prepared media should be brought to room temperature.

  1. Inoculate broth in proportion 1:10 of macerates of biological, foods, faeces and environmental samples.
  2. Incubate aerobically at 30° C for 24-48 hours.
  3. Check for a positive black-brown changed color.
  4. Subculture all broths onto Palcam Agar (#1116).
  5. Incubate at 35° C for 24-48 hours.
  6. Examine for presumptive colonies.
  7. Confirm each presumptive colony by appropriate media and biochemical and serological testing kit.

LIMITATIONS OF METHOD
This medium is only a part of the identification. Other tests may be required.
