PORPHYRIN TEST MEDIUM

Porphyrin Test Medium is used to differentiate Haemophilus sp. on the basis of their ability to synthesize heme (factor X). These Haemophilus may growth without exogenous source of factor X.

PRECAUTIONS
This medium is for IN VITRO diagnostic use only.


STORAGE
Store prepared media at 2-8º C protected from direct light.


SIGN OF DETERIORATION
Media should not be used if the expiry date has passed. Prepared media should not be used if there are signs of contamination or deterioration (evaporation or discoloration).


PROCEDURE
Prior to inoculate, the prepared media should be brought to room temperature.

  1. Inoculate tubes with a suspension of 3-5 similar colonies taken from an overnight growth on Chocolate Agar or Chocolate Bacitracine Agar.
  2. Incubate at 35º C with 5% CO2 for 4-24 hours.
  3. Add an equal volume of Kovac's Reagent (8552). Shake vigourously on vortex.
  4. Let it stand for 2-5 minutes.

Positive results: reddish coloration of the lower part
Negative results: no change in color

LIMITATIONS OF METHOD
Color change will appear in the aqueous-lower phase. A pinkish-red color appearing in the alcohol phase means a negative test result. This medium is only a part of the identification. Other tests may be required.
