Mycological Agar is a solid general purpose medium used for isolation, growth and maintenance of yeasts and molds. Peptone and dextrose are the nutrious agents for yeasts and molds recovery.

PRECAUTIONS
This medium is for IN VITRO diagnostic use only.


STORAGE
Store prepared media at 2-8° C protected from direct light and dehydrated powder, in a dry place, in tightly-sealed containers at 2-25° C.


SIGN OF DETERIORATION
Media should not be used if the expiry date has passed. Prepared media should not be used if there are signs of contamination or deterioration (shrinking, cracking, evaporation or discoloration). Do not use dehydrated media if it is caked.


DIRECTIONS
Suspend 36 g of dehydrated media into 1000 ml of purified filtered water. Heat with frequent agitation and boil for one minute. Dispense into culture tubes and sterilize at 121° C for 15 minutes.


PROCEDURE
Prior to inoculate, the prepared media should be brought to room temperature.

  1. Inoculate tubes with specimens as soon as possible and streak to obtain isolated colonies. Stab cutaneous specimens into surface of the agar. Inoculate in duplicate.
  2. Incubate one set of tubes at 25° C for up to 4-6 weeks and the other set at 35° C for up to 7 days, with loosened caps.
  3. Examine every day or every 2-3 days for growth.
  4. Culture media for strain maintenance should be inoculated with a pure growth and incubate at 25° C for 7 days with loosened caps.

This medium is only a part of identification. Other tests may be required.
