T.S.C. (TRYPTONE SULFITE CYCLOSERINE) AGAR

T.S.C. Agar is a solid selective medium used for isolation and enumeration of sulphite-reducing anaerobes especially Clostridium perfringens, in foodstuffs. The addition of Ferric Ammonium Citrate and Sodium Sulphite enhance the H2S production with blackening of the colonies of Clostridium perfringens D-Cycloserine inhibits growth of others micro-organisms, reduces black halo size to permit a good recovery of sulphite-reducing anaerobes.

PRECAUTIONS
This medium is for laboratory use only.


STORAGE
Store prepared media at 2-8° C protected from direct light and dehydrated powder, in a dry place, in tightly-sealed containers at 2-25° C.


SIGN OF DETERIORATION
Media should not be used if the expiry date has passed. Prepared media should not be used if there are signs of contamination or deterioration. Do not use dehydrated media if it is caked.


DIRECTIONS
Suspend 42,0 g of dehydrated media in 1000 ml of purified filtered water. Heat with frequent agitation and boil for one minute. Sterilize at 121° C for 15 minutes. Cool down to 45° C and add 2x5 ml of D-Cycloserine Supplement (#8749). Mix well and dispense into sterile Petri plates.


PROCEDURE
Prior to inoculate, the prepared media should be brought to room temperature.

  1. Inoculate specimens directly and streak for colony’s isolation
  2. Incubate at 35° C for 24 hours under anaerobe’s atmosphere.

LIMITATIONS OF METHOD
